- Diese Arbeit verfolgt das Ziel, den extrazellulären Elektronentransferweg des extrem acidophilen Bakteriums Acidithiobacillus ferrooxidans durch Einbringen der Proteinkaskade in Escherichia coli und Vibrio natriegens zu charakterisieren. Die E. coli Stämme BL21(DE3), T7 Express und BL21 Gold(DE3) DeltaABCF konnten kein Holo-CycA produzieren und die löslichen periplasmatischen Proteine waren schwach an die Zellmembranen assoziiert. Dagegen waren V. natriegens Vmax X2 und E. coli C43(DE3) in der Lage, alle vier Redox-Proteine im korrekten Zellkompartment zu produzieren und die c-Typ Cytochrome waren redox-aktiv. Der neue E. coli C43(DE3) Stamm reduzierte doppelt so viel Fe(III)-Citrat und die Stromproduktion in einer mikrobiologischen Brennstoffzelle (MBZ) war fünf Mal höher im Vergleich zur Wildtyp-Kontrolle. Der rekombinante Elektronentransferweg in E. coli C43(DE3) war redox-aktiv unter Umsatzbedingungen in der MBZ. Dies ist die erste Demonstration einer funktionellen rekombinanten Proteinkaskade des extrem acidophilen Bakteriums At. ferrooxidans in E. coli.
- Abstract (EN)
- This work aims at characterizing the extracellular electron transfer (EET) pathway of the extreme acidophile Acidithiobacillus ferrooxidans by introducing the proposed protein cascade into Escherichia coli and Vibrio natriegens. The E. coli strains BL21(DE3), T7 Express, and BL21 Gold(DE3) DeltaABCF were unable to produce holo-CycA and the soluble periplasmic proteins were weakly associated to the membranes. In contrast, V. natriegens Vmax X2 and E. coli C43(DE3) were able to produce all four redox proteins in the correct cell compartment and the c-type cytochromes were redox active in vitro. The new E. coli C43(DE3) strain reduced two times more Fe(III)-citrate and current production in a microbial fuel cell (MFC) was increased 5-fold compared to the wildtype control. The recombinant EET pathway in E. coli C43(DE3) was redox active under turnover conditions in the MFC. To the author’s knowledge this is the first demonstration of a functional recombinant redox protein cascade from the extreme acidophile At. ferrooxidans in E. coli.
